Passive and active (Pgp-mediated) cellular transport, along with plasma protein binding, are key determinants of a compound’s ability to traverse the blood-brain barrier into the CNS. High-quality in vitro measurements of these properties, combined with novel integrative models are used to prioritize compounds for in vivo studies of CNS penetration.
